Ramanaidu Daggubati was born to Telugu cinema producer D. Suresh Babu in Madras (now Chennai), Tamil Nadu. He is named after his paternal grandpa, Telugu cinema producer D. Ramanaidu.
His paternal uncle Venkatesh and cousin Naga Chaitanya are also actors, and he is a part of the Daggubati–Akkineni family.
Rana disclosed in 2016 that he is blind in one eye and has a transplanted left eye. The surgery was performed at Hyderabad’s L. V. Prasad Hospital. When he was 14, he had another operation on his right eye, but it failed.
