Rama Rao was born on May 20, 1983, to Nandamuri Harikrishna and Shalini Bhaskar Rao, a film actor and politician.

His father is of Telugu ancestry, while his mother is a Kannada Brahmin from Kundapur, Karnataka.
He is the grandson of Telugu actor and former Andhra Pradesh Chief Minister N. T. Rama Rao. On his grandfather’s advice, he was renamed N. T. Rama Rao after his first name, Tarak.

At the age of seven, Rama Rao made his film debut as a child actor in Brahmarshi Vishwamitra (1991). He played Bharata in the film written and directed by his grandfather, N. T. Rama Rao.

However, the film went into production for much too long. Producer Ramoji Rao later signed him and he chose to work on the romance drama Ninnu Choodalani, which was his first lead role.
He was only 17 at the time. Student No. 1, which came out later, was a commercial triumph, whilst Subbu (2001) was a flop.

For the third time, Rama Rao worked with filmmaker Rajamouli on the socio-fantasy film Yamadonga. He had to lose more than 20 kg for this film since he used to weigh 94 kilograms.

Rama Rao married Lakshmi Pranathi, the daughter of Narne Srinivasa Rao, a realtor and businessman. N. Chandrababu Naidu, who mediated the marriage, is Srinivasa Rao’s niece.
Their wedding was placed at the Hitex Exhibition Center in Madhapur on May 5, 2011. They have two sons together.
